| 目的 研究大肠癌中增殖细胞核抗原PCNA、p53、nm23的表达及临床病理特征间的关系。方法 应用SP免疫组织化学方法检测77例大肠癌中PCNA、p53、和nm23的表达。结果 PCNA表达强弱与肿块大小、分化程度、浸润深度、淋巴结转移及临床分期密切相关;p53与淋巴结转移及临床分期密切相关;nm23表达与淋巴结转移有统计学意义。结论 PCNA与p53蛋白的表达强度与大肠癌的进程呈正相关,nm23的强表达与淋巴结转移负相关,PCNA与p53正相关,与nm23负相关。由此可见,三项指标的综合表达对大肠癌的临床诊治及其预后评估有重要意义。
The expression of proliferating cell nuclear antigen,p53,nm23 and their significance in human colorectal carcinoma

Abstract:Objective To understand the relation between the expression of PCNA,p53,nm23 and the clinical pathology in human colorectal carcinoma. Method The expression of PCNA,p53 and nm23 in seventy-seven human colorectal carcinomas was studied with sp immunohistochemical technique. Results The expression of PCNA,p53,nm23 in the tumor is correlated with lymph node metastasis and clinical stage.Correlation was also observable in the expression of PCNA and the tumor size,infiltration depth. Conclusion The expression of PCNA and p53 protein are positively correlated to the cancer development,but nm23 has a negative correlation to the metastasis.So the three markers have important significance in clinic treatment and prognosis for colorectal carcinoma.

增殖细胞核抗原PCNA与抑癌基因p53蛋白在多种肿瘤中均有表达,且与肿瘤分化、转移、预后均相关,肿瘤转移抑制基因nm23的表达与淋巴结转移相关,但在大肠癌组织内尚无定论[1,2]。本文检测PCNA、p53、nm23在原发性大肠癌中的表达情况,评价以上三项免疫指标在大肠癌的分类诊断及其临床预后判断中的意义。
材料和方法
一、选取本院1996年6月~1998年6月77例大肠癌手术切除标本,男47例,女20例,所有患者术前均未经化疗或/和放疗,根据《中国常见肿瘤的诊治规范》的标准进行分类 。本组77例中,高分化腺癌17例,中分化腺癌34例,低分化腺癌9例,粘液腺癌17例。每例在肿瘤中央及边缘取组织三块,正常肠粘膜3例,经10%的中性福尔马林固定,常规石蜡包埋,连续切片,HE染色。
